Custom Cell Line Creation
Our custom cell line labeling services consist of developing “labeled” recombinant bioluminescent or fluorescent cell lines of interest to generate relevant non-invasive animal models. The parental cell line is typically commercially available from the Caliper Bioware portfolio or is supplied by our clients or can be chosen from the CDAS in-house panel of over 200 human cancer cell lines. The experimental procedure consists of two steps with the creation of the recombinant cell line and its characterization to ensure that the newly created line retains the properties of the parental line.
- Cell line creation
CDAS utilizes a specific lentiviral-based system allowing transformation of slow and fast dividing cell lines with a very high efficiency. Recombinant cell populations are then enriched and recombinant luciferase clones are isolated. Luciferase activity is measured in order to functionally analyze the cells during the process of clone selection and amplification.
- Cell line characterization
- Light emission is measured and provided in photons/second per cell (minimum of 100 photons per cell per second)
- In vitro growth rate is compared to that of the parental cell line
- Cell lines are routinely tested to ensure they are bacteria and mycoplasma-free
Additional luciferase cell line characterization is offered to determine:
- The stability of the light emission for 4 weeks in the absence of the selective pressure
- The in vitro response to one standard drug compared to the parental cell line response
- The in vivo tumorigenicity is compared to the parental cell line in a subcutaneous efficacy testing with a single response to one standard drug
